Roche RTS ProteoMaster Instrument. The RTS ProteoMaster Instrument, designed for use with RTS 100, RTS 500 and RTS 9000 kits, provides optimal protein expression conditions by regulating the reaction temperature and by smooth shaking that supports the continuous exchange between reaction and feeding chamber. Temperature is controlled by circulating air that surrounds the reaction devices from multiple sides, thereby preventing condensation on the lids of the devices. It supports the incubation of all RTS reaction formats. There is an optimized tool for scale up to preparative levels. Inserts for different reaction formats are provided. Reactions are carried out in the new RTS 500 Reaction Device. This device is composed of a reaction compartment and a feeding compartment. The vertical reaction compartment, where the coupled transcription/translation reaction takes place, is separated from the feeding compartment by two semipermeable membranes. The plasmid containing the gene of interest serves as a template for transcription. The 10-ml feeding compartment supplies substrates through the semipermeable membranes. By-products that may inhibit the reaction are simultaneously diluted via the same membranes into the feeding compartment. This continuous exchange of substrates and by-products across the two semipermeable membranes (CECF technology) allows protein synthesis to proceed for up to 24 hours.
